Genetic Alterations
These mice carry genetic variations in the progesterone receptor. The following nucleic acid alterations were introduced, which predicts amino acid substitutions at codons 461, 478 and 489 from KKR (lysine, lysine, arginine) to FFF (phenylalanine, phenylalanine, phenylalanine). More specifically, K461F: AAA to TTT, K478F: AAG to TTT, and R489F: CGG to TTC.

The progesterone receptor (Pgr) is a nuclear steroid receptor that functions as a ligand activated transcription factor regulating gene expression following progesterone hormone binding. The progesterone receptor gene has two distinct promoters and translation start sites in the first exon which produce two isoforms, A and B. PgR_AF1_FFF knock-in mice carry K461F, K478F and R489F (lysine, lysine, arginine, respectively to phenylalanine) substitutions in the activation function 1 domain of Pgr. Males homozygous for the mutation are viable and fertile. Female PgR_AF1_FFF mice are infertile and exhibit impaired mammary and uterine development. In cell cultures the FFF mutations result in the abolition of up to 80% of progesterone receptor activity. This strain may be useful for studying the biology of the mammary gland and uterus.

PgR_AF1_QQQ mice carrying the K461Q, K478Q and R489Q mutations are available as MMRRC Stock No. 067159.

Strain Development
CRISPR/Cas9 endonuclease-mediated genome editing was used to create an amino acid substitutions at positions 461 (lysine to phenylalanine, K461F, AAA to TTT), 478 (lysine to phenylalanine, K478F, AAG to TTT), and 489 (arginine to phenylalanine, R489F, CGG to TTC) in the Pgr (progesterone receptor) gene. Single guide RNA and Cas9 endonuclease were introduced into C57BL/6J-derived fertilized eggs by pronuclear injection. Embryos were transferred to pseudopregnant females, and correctly targeted pups were bred to C57BL/6J mice for germline transmission. The mutation was confirmed by sequencing. The colony was maintained on the C57BL/6J background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
