Strain Development
The Y BAC transgenic lines were produced originally by Paul Burgoyne (Francis Crick Institute, formerly National Institute for Medical Research, London) and Michael Mitchell (Aix Marseille Université, France), as part of their studies of Y genes that are critical for spermatogenesis. Classical methods of gene targeting were unsuccessful for Y genes, and thus Burgoyne and Mitchell used deletion mutants to find small regions of the Y chromosome that contain genes critical for spermatogenesis. Then, they added back Y genes as BAC transgenes, one by one, to determine which Y genes rescued the deficit in Y chromosome deletion variants with defined spermatogenic failure (e.g., PMID:11528390 and 21530259]). The Eif2s3x line was produced by Monika Ward at the University of Hawaii (PMID:24263135), who made the transgenic insertion using a BAC construct produced by Michael Mitchell. The 7 BAC transgenic lines were sent to Art Arnold (UCLA) on an MF1 or mixed MF1 background beginning in 2012, who backcrossed them for 10 or more generations to C57BL/6J. The Eif2s3x line was also backcrossed to MF1 at UCLA.

BAC DNA was injected as closed circles into fertilized eggs from CBA/Ca × C57BL/10J or randomly bred MF1 matings at the National Institute for Medical Research (PMID:21530259). The transgenic founders were crossed by the Burgoyne Lab to MF1 random-bred albino females, using BAC-specific primers listed below. In many cases, the copy number of the transgene was assessed by Southern-blot analysis (e.g., PMID:21093264). In cases where no single BAC encoded the entire gene, recombineering was used to add the missing exons as spliced cDNA.
