Not published but the following information was provided by the donating investigators laboratory:

  • C2 and D4 lines were derived during the same experiment in 1999. These two lines had best chromosome count among others and were determined XY by PCR at that time (which does not preclude them from loosing Y chromosome during in vitro culture). Both C2 and D4 aggregated at early passage with outbred ICR morulae and resulted in chimeras that did not, however, transmit through germline.
  • Both lines were tested for chimera generation before any genetic manipulations by a collaborator who found that using injections with BALB/c blastocysts produced germline chimeras. Prolonged in vitro culture during gene targeting found D4 to be more susceptible to differentiation than C2. This collaborator has now used C2 for quite a few years and has satisfactory recombination and generation of chimeras with BALB/c blastocysts.
  • Donor did not test generation of chimeras using BALB/c or albino C57BL/6 host embryos in his lab. Donor subcloned both C2 and D4 lines in TX medium manufactured by Belgian company Thromb-X (now distributed by Chemicon under the name of RESGRO). After subcloning in TX medium, both lines were capable of generating germline transmitting chimeras after aggregations with ICR embryos.
  • Donor performed one electroporation in his lab using the C2 line, two subclones with randomly integrated transgene (EGFP) have produced germline transmitting chimeras after aggregations with ICR embryos. TX-subcloned pool of C2 were used for electroporation in regular medium. After identifying good expressors, cells were grown in TX-medium for two passages before aggregations.
