Genetic Alterations
Cnga3tm1Biel: The Cnga3 gene was disrupted by replacement of exon 7 with a neomycin resistance cassette. Absence of gene expression in homozygous mutant animals was confirmed by RT-PCR analysis of retina RNA.

Rhotm1Jlem: A PGK-neo cassette was inserted into the first coding exon of the endogenous gene, deleting 15 bp upstream of the translational start site and 111 codons. Transcript was undetected in homozygous mutant mice by Northern blot and RT-PCR analyses. Western blot analysis indicated an absence of protein in whole retinal homogenates.

Phenotype
B6J.129-Rhotm1JlemCnga3tm1Biel/LrdrMmjax (Cnga3-/-; Rho-/-), coneless and rodless double knockout mice, show no pupil response to low and medium-intensity red light. Pupil responses to other stimuli revealed a melanopsin-mediated contribution; however, a 40–60% reduction in maximal constriction amplitude indicated that rods, cones, and melanopsin all contribute to normal pupil constriction in wild-type mice. Similar to single knockout models, Cnga3-/-; Rho-/- mice showed an increased sust/max ratio to blue stimuli, indicating delayed pupil dilation in the later postillumination phase due to loss of classic photoreceptor function. Furthermore, a prolonged constriction to high-intensity red light is observed, likely driven by melanopsin activation. At one month of age, no rod nor cone function is detected by electroretinogram nor pupillometry in the Cnga3-/-; Rho-/- mice.

This model provides a valuable opportunity to study a retina that lacks rod and cone function, but still exhibits inner retinal photosensitivity,

Strain Development
Cnga3tm1Biel: Cng3 DNA was isolated from a genomic P1 library made from 129/Sv mouse tissue. Three exons were identified corresponding to exons 5–7 of the CNG3 gene. A targeting vector was constructed and the herpes simplex thymidine kinase cassette was cloned 39 to the CNG3 sequence. The targeting vector was linearized and electroporated into R1 ES cells. G418/gancyclovir-resistant clones were screened by Southern blot analysis, and ES cells carrying the disrupted allele were microinjected into C57BL/6 blastocysts to produce chimeric mice. Chimeric mice were crossed with strains C57BL/6 (outbred) and 129/Sv (inbred). Heterozygotes then were intercrossed to produce mice homozygous for the CNG3 mutation.

Rhotm1Jlem: A targeting construct was made from a 129/Sv genomic clone. J1 embryonic stem cells were electroporated with the targeting construct and selected in G418 and gancyclovir. Of the 79 clones, 13 were identified as homologous recombinants via EcoRI digestion. In addition, six putative recombinants were verified by BamHI digestion.

Cnga3tm1Biel and Rhotm1Jlem were crossed and then backcrossed with C57BL/6J for at least 5 generations.
