Genetic Alterations
ENU-induced A to G transition at base pair 3,436,475 (v38) on chromosome 2, or base pair 12,684 in the GenBank genomic region NC_000068 for the Dclre1c gene. The mutation corresponds to residue 431 in the mRNA sequences NM_146114, NM_175683, NM_001110214 within exon 5 of 14, 16, and 15 total exons, respectively. The mutation results in a histidine (H) to arginine (R) substitution at position 115 (H115R) in the Artemis protein.
Phenotype

Homozygous: Mice showed a decreased frequency of B cells and a decreased frequency of T cells including both CD4+ and CD8+ T cells as well as an increased frequency of macrophages, neutrophils, and natural killer (NK) cells, all in the peripheral blood. The T-independent antibody response to 4-hydroxy-3-nitrophenylacetyl-Ficoll (NP-Ficoll) was also diminished.

Heterozygous: No signficant abnormal phenotype detected.

Original mutant was a C57BL/6J G3 ENU-induced mutant; all subsequent crosses to maintain strain were on C57BL/6J background only.
