ENU-induced T to C transition at base pair 66,841,172 (v38) on chromosome 4, or base pair 13,362 in the GenBank genomic region NC_000070 for the Tlr4 gene. The mutation corresponds to residue 2,482 in the mRNA sequence NM_021297 within exon 3 of 3 total exons. The mutation results in a valine (V) to alanine (A) substitution at position 734 (V734A) in the TLR4 protein.

Strain Description

Phenotype

Homozygous: Mice were deficient in TNFα secretion from macrophages in response to the Toll-like receptor 4 (TLR4) ligand, lipolysaccharide (LPS). A mouse in the pedigree also exhibited reduced B cell frequency including IgD+ B cells and IgM+ B cells, all in the peripheral blood.

Heterozygous: No abnormal phenotype detected.
