Genetic Alterations
ENU-induced T to C transition at base pair 155,172,561 (v38) on chromosome 2, or base pair 39,183 in the GenBank genomic region NC_000068 for the Itch gene, corresponding to residue 520 in the mRNA sequence NM_008395 (variant 1) within exon 5 of 25 total exons and residue 722 in the mRNA sequence NM_001243712 (variant 2) within exon 6 of 26 total exons. The mutation results in a leucine (L) to serine (S) substitution at position 106 (L106S) in both variants of the Itch protein.
Phenotype

Homozygous: Mice exhibited increased total IgE levels in the peripheral blood and an enhanced IgE response to the cysteine protease allergen, papain.

Heterozygous: No abnormal phenotype detected.
