Genetic Alterations

A loxP site was inserted upstream of exon 9 (ENSMUSE00001293030). An FRT-flanked neomycin resistance cassette with a 5' loxP site was inserted downstream of exon 9. Flp-mediated recombination removed the selection cassette. Cre-mediated recombination removed exon 9.

Phenotype
Epac2 knockout mice were born at the expected Mendelian ratios, viable and fertile, and developmentally normal without obvious abnormality. Epac2-/- mice developed polyuria (PMID:30252533).

Strain Development
R1 embryonic stem cells derived from (129X1/SvJ x 129S1/Sv)F1-Kitl+ were used for homologous recombination and implanted into C57BL/6 blastocysts. Male chimeras were mated with female black Swiss mice. The selection cassette was deleted by mating to B6.Cg-Tg(ACTFLPe)9205Dym/J (RRID:IMSR_JAX:005703), and exon 9 was excised by breeding to 129S/Sv-Tg(Prm-cre)58Og/J (RRID:IMSR_JAX:003328, which was crossed with C57BL/6 for >5 generations). Subsequent backcrossing to C57BL/6NCrl for >20 generations.
