Genetic Alterations
A targeting vector was designed to insert a mutant exon 5 containing a deletion of trinucleotide GAA corresponding to the human codon 302. The cassette also carried a PGK-neo-pA-STOP, with a false translation signal, splice donor site and another polyA tail inserted into intron 4. Crossing with mice expressing cre in the germline removed the PGK-neo-STOP cassette. Heterozygous mice expressed transcript from the mutant allele.

Phenotype

Tor1atm1Yql mice exhibit an age and sex-dependent phenotype. Homozygous mutants are embryonic lethal, while heterozygotes are viable with normal brain anatomy. Heterozygous mice displayed deficient performance on the beam-walking test, hyperactivity in the open-field test, and a gait abnormality of increased overlap. Mice at 3 months of age did not display deficits in beam-walking and gait, while 6-month mutant mice did, indicating an age factor in phenotypic expression.

While striatal dopamine and 4-dihydroxyphenylacetic acid (DOPAC) levels in Tor1atm1Yql mice were similar to that of wild-type mice, a 27% decrease in 4-hydroxy, 3-methoxyphenacetic acid (homovanillic acid) was detected in mutant mice. Tor1atm1Yql tissues also have ubiquitin- and torsinA-containing aggregates in neurons of the pontine nuclei.

A sex difference was noticed in the mutant mice with female mutant mice exhibiting fewer alterations in behavioral, neurochemical, and cellular changes. Our results show that knocking in a Tor1atm1Yql allele in mouse alters their motor behavior and recapitulates the production of protein aggregates that are seen in dystonic patients. The data further support alterations in the dopaminergic system as a part of dystonia’s neuropathology.

Background Genetic Quality:

The MMRRC has developed a Genetic Quality Control pipeline using the MiniMUGA array to provide additional information to identify and validate genetic backgrounds of MMRRC strains. For more information on whether genetic background data is available, please contact MMRRC_GeneticQC@med.unc.edu. Note: that MiniMUGA genetic background data is not available on all strains, but can be ordered if desired.

Strain Development
The Tor1a (Dyt1) gene of 129/Sv origin used for targeting vector construction came from a Bacterial Artificial Chromosome Mouse I library, screened by Incyte Genomics, Inc. (clone addresses 114/N12 and 168/J4). Exon 5 of the gene was mutagenized by PCR to carry a deletion of trinucleotide GAA. In mouse, the DNA sequence that codes for glutamic acid pair at the position corresponding to human exon 302/303 is GAAGAG rather than GAGGAG. GAA is a synonymous codon of GAG; both code for the glutamic acid residue. The donating investigator removed the GAA to leave behind a GAG that is homologous with the remaining GAG in DYT1 sequence of dystonic patients. In addition to the trinucleotide deletion, the construct had a cassette carrying PGK-neomycin, poly(A), and a STOP sequence with a false translation signal, splice donor site, and an additional poly(A) tail inserted into intron 4. The targeting construct was linearized and transfected into D3 129/SvJ mouse embryonic stem (ES) cells. After selection with G418 antibiotics, colonies were isolated and screened by Southern analysis for homologous recombination for both sides. PCR was done to detect the presence of the deletion. Colonies with the targeted allele without GAG were expanded and injected into C57BL/6 blastocysts. Chimeric offsprings were crossed with C57BL/6 mice to obtain germline transmitted pups. To produce the Dyt1 knockin mice, heterozygous germline transmitted mice were mated with CMV-cre mice to remove the PGKneoSTOP sequence. Double positive heterozygotes were mated with wild-type mice to generate a group of test animals containing wild-type animals and single heterozygotes containing the mutated Dyt1. Mice were genotyped using a pair of primers specific to the sequence flanking the insertion site and a pair specific to cre. To confirm that one allele of the mutant mouse carries the mutation, the mRNA of the region surrounding the mutation site was sequenced. Whole brain RNA was isolated from adult wild-type and Dyt1 knockin mice using Qiagen RNeasy Protect Kit. RT-PCR was performed using materials and protocols provided by Qiagen OneStep RT-PCR.

Disclaimer: If MMRRC Strain Genetic Quality Control (GQC; based on MiniMUGA genotyping and analysis) has been completed for this strain, the information might differ from the genetic background information provided by the submitter. MiniMUGA genetic analysis is done on a strain's tissue samples taken when archived by or ordered from the assigned MMRRC Center.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter.
