4 Recovery refers to thawing, in vitro fertilization (IVF), and/or embryo culture leading to live offspring.

Emx1tm1(cre)Yql
Name: empty spiracles homeobox 1; targeted mutation 1, Yuqing Li
Type: Allele
Species: Mus musculus (mouse)
Chromosome: 6
Alteration at locus: Knock-In
Emx1
Name: empty spiracles homeobox 1
Type: Gene
Species: Mouse
Chromosome: 6
NCBI: 13796
HGNC: HGNC:3340
Homologene: 55799
Genetic Alterations
A knock-in strategy was used to express cre in an EMX1- specific manner. A lacZ-IRES-cre cassette replaced a segment of the gene containing the first four amino acids of the encoded protein. This allele expresses a bicistronic mRNA encoding beta-galactosidase and Cre recombinase. The endogenous promotor is active in the cerebral cortex and hippocampus.

ES Cell Line
D3
Phenotype
Emx1 knock-in mice are normal and show no overt phenotype. Beginning around embryonic day 9.5 (E9.5), heterozygous Emx1-cre knock-in mice express the Emx1 gene in virtually every neuron of the developing embryonic and postnatal cerebral cortex. Regionally, Emx1 is also expressed in postmitotic neurons that will later form different layers of the adult cortex. Cre expression is functional, mediating loxP-dependent recombination in cortical tissue. This line should be conducive to further studies of the effect of a gene mutation or overexpression upon the development and plasticity of cerebral cortex and hippocampus

Strain GQC Summary
Gene Specific Genotyping:

To request gene-specific and other genotyping services for a strain, please contact the distribution MMRRC Center for more information.

Background Genetic Quality:

The MMRRC has developed a Genetic Quality Control pipeline using the MiniMUGA array to provide additional information to identify and validate genetic backgrounds of MMRRC strains. For more information on whether genetic background data is available, please contact MMRRC_GeneticQC@med.unc.edu. Note: that MiniMUGA genetic background data is not available on all strains, but can be ordered if desired.

Strain Development
A targeting construct was developed to delete the 18-bp NotI–SacII fragment encoding the first 4 amino acids of the Emx1 protein and an insert a 7 kb fragment containing a coding region for a nuclear-localized b-galactosidase fused in frame with the Emx1 coding region, IRES, a coding region for a nuclear-localized Cre protein, SV-40 poly(A) sequence, and a PGKneo-poly(A) cassette. D3 ES cells were cultured and electroporated and resistant clones were isolated following 7 days of G418 selection. Targeted ES cell clones were expanded and chimeric animals were produced. On average, 10 to 15 ES cells were injected into the C57BL/6 blastocysts. The injected blastocysts were cultured and inserted into the uteri of foster mothers. Chimeric animals were mated with C57BL/6 or F1B6DBA2 animals to test for germline transmission. The resulting F1 and F2 animals were were backcrossed to C57BL6/J for more than 6 generations. Genotypes of all offspring were analyzed by either PCR or Southern blot.

Disclaimer: If MMRRC Strain Genetic Quality Control (GQC; based on MiniMUGA genotyping and analysis) has been completed for this strain, the information might differ from the genetic background information provided by the submitter. MiniMUGA genetic analysis is done on a strain's tissue samples taken when archived by or ordered from the assigned MMRRC Center.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter.
