Genetic Alterations
Kmt2dAID mice express an AID-tagged Kmt2d allele; these mice may be used in the second-generation auxin-inducible degron (AID2) system. When combined with an OsTir1-F74G-expressing mouse line or viral vector, the resulting progeny will demonstrate rapid and reversible degradation of KMT2D-AID tagged protein after exposure to the auxin derivative 5-Ph-IAA.

Lysine (K)-specific methyltransferase 2D (coded by Kmt2d), also referred to as MLL4, is widely expressed in adult tissues and is essential for in regulating development, differentiation, metabolism, and tumor suppression. Mutations in Kmt2d are associated with developmental diseases such as Kabuki syndrome and congenital heart disease, and various forms of cancer.

These Kmt2dAID mice have an AID tag inserted immediately prior to the start codon of the Kmt2d gene and may be used in the second-generation auxin-inducible degron (AID2) system. When combined with a mouse line, or viral vector, that expresses the Japanese rice Oryza sativa F-box transport inhibitor response 1 (TIR1, formal name LOC4337827) protein, harboring the F74G modification, the resulting progeny will demonstrate rapid and reversible degradation of KMT2D-AID tagged protein, after exposure to the auxin derivative 5-phenyl-indole-3-acetic acid (5-Ph-IAA).

Homozygous mice are viable and fertile. The donating laboratory bred these Kmt2dAID mice with a Kmt2cf; Cre-ER conditional mouse line and primary preadipocytes were obtained from the resulting Kmt2cf/f; Kmt2dAID/AID; Cre-ER (RRID:MMRRC_075967-JAX) progeny. After exposure to tamoxifen, the Cre-recombined Kmt2c null; Kmt2dAID cells were then infected with retroviruses expressing Oryza sativa TIR1, harboring the F74G modification. Depletion of KMT2D in the cells occurred within 2hours after treatment with 5-Ph-IAA. These Kmt2dAID mice may be useful for studying the role of the KMT2D protein in cell differentiation and organogenesis.

Strain Development
The Kmt2dAID allele was generated using CRISPR/Cas9 endonuclease-mediated genome editing. A single guide RNA (5'-TGTCCATCCCTCTCTCCGAC-3') was used to insert a minimal auxin-inducible degron sequence after the start codon located on exon 2 of the lysine (K)-specific methyltransferase 2D (Kmt2d) gene on chromosome 15 (ENSG00000167548). The sgRNA, ssDNA, and the Cas9 nuclease were introduced into the cytoplasm of zygotes derived from B6D2F1/J mice with well recognized pronuclei. Correctly targeted pups were identified by PCR and sequencing. The resulting Kmt2dAID mice were backcrossed to C57BL/6J mice for at least 5 generations by the donating laboratory. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
