Phenotype
Mice do not exhibit an overt phenotype that differs from wildtype. However, following tamoxifen-induced Cre recombination (deletion of Kmt2c) and auxin-mediated depletion of Kmt2d, cells display severe defects in enhancer function, including impaired de novo binding of the lineage-determining transcription factor MyoD and reduced recruitment of chromatin modifiers BAF and p300 to enhancers. Acute loss of Kmt2c and Kmt2d disrupts enhancer activation marked by reduced H3K4me1 and H3K27ac, leading to compromised chromatin accessibility and defective transcriptional activation during myogenic reprogramming.

Strain Development

Kmt2ctm1.2Jwle: A loxP site was inserted into intron 56 and an FRT site flanked neomycin resistance gene cassette followed by a second loxP site was inserted into intron 58. Sequence encoding 61 of the 121 amino acids of the SET domain were thus enclosed within loxP sites. The neo cassette was removed through subsequent Flp-mediated recombination, leaving exons 57-58, including sequence encoding the RYINHS motif of the catalytic core region of the SET domain, flanked by loxP sites.

Gt(ROSA)26Sortm1(cre/ERT2)Tyj: A cassette containing Cre-ERT2 was inserted into the Gt(ROSA)26Sor locus.

Kmt2dem2Kaig: Using CRISPR/Cas9 endonuclease-mediated genome editing, a single guide RNA (5'-TGTCCATCCCTCTCTCCGAC-3') was used to insert a 132bp DNA fragment encoding an Auxin-Inducible Degron (AID) tag sequence after the start codon located on exon 2 of the lysine (K)-specific methyltransferase 2D (Kmt2d) gene.

Successful integration of the AID tag was confirmed with genotyping and Sanger DNA sequencing. Kmt2dAID/AID mice were crossed with Kmt2c f/f (Kmt2ctm1.2Jwle); Cre-ER mice (Gt(ROSA)26Sortm1(cre/ERT2)Tyj) to obtain Kmt2c f/f; Kmt2dAID/AID; Cre-ER mice.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
