Genetic Alterations

Using CRISPR/Cas9 endonuclease-mediated genome editing. A single guide RNA (5'-GCAAAGGACGGGCAAGCGGA) was used to introduce a conditional by inversion (COIN) cassette into intron 1 of the luteinizing hormone/choriogonadotropin receptor (Lhcgr) gene. The entire construct has an initial inward facing lox2722 site, an inward facing lox5171 site, the inverted cassette containing a rabbit beta-globin splice acceptor (SA), a P2A self-cleaving peptide, an enhanced green fluorescent protein gene (EGFP), and a bovine growth hormone (bGH) polyadenylation signal. The inversion cassette is then followed by lox2272 and lox5171 sites, both in reverse orientation.

Phenotype

Lhr-COIN mice do not display any overt phenotypes under baseline conditions.

Conditional Phenotype:
Following Cre-mediated inversion of the COIN cassette, homozygous Lhr inverted mice lack Lhr expression in the DNA and mRNA. Mice with the inverted allele are infertile and have smaller reproductive organs.

Strain Development
This Lhcgr conditional knockout strain was developed via CRISPR/Cas9-mediated genome editing. CRISPR guides, Cas9 protein, and the donor vector were microinjected into C57BL/6J zygotes. Progeny were screened for the desired mutation and founder mice were mated to C57BL/6J breeders (RRID:IMSR_JAX:000664). N1 progeny were identified via PCR and sequencing and then mated again to C57BL/6J breeders or other N1 mice to generate homozgyous mice.

If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
