Phenotype
Homozygous: Mice that are homozygous for the targeted mutation are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Stimulated CD4+ T cells exhibit increased interferon gamma production in vivo using low anti-CD3 concentrations; stimulated CD8+ T cells exhibit increased interferon gamma production, but only at higher antibody concentrations, in both cases, T cell proliferation was not effected. OVA primed CD8+ T cells produce 5-10 fold more interferon gamma than controls. T cell clonal expansion is augmented and cytolysis is increased in mutant antigen-specific CD8+ T cells. Unlike experimental immune encephalitis (EAE) resistant 129S4/SvJae mice, immunized mutant mice on the 129S4/SvJae background develop severe EAE with early onset and rapid progression. This mutant mouse strain may be useful in studies of T cell activation, tolerance and susceptibility to autoimmune disease.

Strain Development
A targeting vector containing neomycin resistance and herpes simplex virus thymidine kinase genes was used to replace the signal exon and two thirds of the IgV exon. The construct was electroporated into 129S4/SvJae derived J1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to 129S4/SvJae mice and heterozygotes interbred. Upon arrival, mice were bred to 129S4/SvJaeJ for at least 1 generation to establish the colony.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
