This allele harbors a targeted knock-in of inverted flip-excision (FLEx) and Foxl2 containing oncogenic C130W amino acid substitution. This mutation is analogous to the human mutation associated with adult type ovarian granulosa cell tumors.

Conditional phenotype: Yes. Upon crossing to Amhr2-Cre, the mouse model recapitulates key aspects of the human adult-type granulosa cell tumors. Including a dependence upon the causative FOXL2 mutation, concomitant uterine hyperplasia, and an age dependent onset of tumors at the time of decreasing ovarian function.

Strain Development
The human FOXL2 p.C134W mutation (c.C402G) corresponds to murine p.C130W mutation by homology. A “flip-excision” (FLEx) switch configuration was usedc in which the targeting construct contained an inverted Foxl2C130W coding sequence flanked by tandem loxP and lox2272 recombination sites. In the presence of Cre recombinase, the Foxl2C130W coding sequence is inverted to restore expression. Homology arms were isolated from C57BL/6J mouse genomic DNA and used to create the targeting vector, which was then introduced into C57BL/6J-693 embryonic stem (ES) cells (derived from C57BL/6J) by electroporation. ES cell clones were then verified using PCR and DNA sequencing to confirm the occurrence of homologous recombination. Recombined ES cell clones were injected into blastocysts, which were then implanted in pseudo-pregnant females to form chimeras. In vivo excision of the neomycin cassette was performed by crossing to a line expressing Flp recombinase, and germline transmission of the final inducible Foxl2C130W-FLEx allele was confirmed by the production of founder animals.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
