Genetic Alterations
A neomycin resistance cassette replaced sequences including the last exon, which encodes the carboxy terminal end of the kinase domain and the cytoplasmic tail. RT-PCR analysis demonstrated that a mutant transcript was expressed from this allele that lacked sequences encoding the cytoplasmic kinase domain.

    Phenotype

    The mice display normal breeding patterns and become blind at approximately 4 months of age. The mice are known to have defective clearance of apoptotic cells and altered immune responses. However, other phenotypes reported earlier in mertk null mixed background mice such as, hyperimmunity, appear to have diminished.

    Strain Development
    A 574 bp cDNA mertk fragment was used to screen a 129/Sv genomic library. The tyrosine kinase domain in Mertk cytoplasmic tail was replaced with neomycin resistant gene. 129P2/OlaHsd E14TG2a embryonic cell line was targeted and resulting mice were bred initially to (C57BL/6 x DBA/2 )F1. The targeting resulted in unstable mRNA and lack of the Mertk protein. Mertk null mice were subsequently backcrossed to C57BL/6J for at least 12 generations and maintained by sibling matings.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
