This strain harbors a CRISPR/Cas9-mediated targeted modification of the Fpr2 gene, replacing the mouse Fpr2 coding sequence with the human FPR2 (ALX) coding sequence.

Phenotype
Homozygous mice do not exhibit any overt abnormalities under baseline conditions.

Following lipopolysaccharide (LPS) saccharide, mice carrying the hFPR2 display features of acute sialadenitis as shown by increased levels of proliferating inflammatory cells, loss of epithelial integrity and reduced saliva secretion.

Treatment with with aspirin-triggered resolvin D1 (AT-RvD1), reduces these features, with decreased inflammatory cell proliferation and restoration of saliva secretion to levels comparable to phosphate buffered saline (PBS)-treated healthy controls.

The changes observed in mice carrying the hFPR2 and treated with LPS and AT-RvD1 were comparable to those observed in wild-type mice carrying the mouse Fpr2.

Strain Development
The Fpr2em1Olgab knock-in mouse line was generated using a DNA repair template designed to insert the human FPR2 coding sequence in place of the corresponding mouse Fpr2 gene. C57BL/6J embryos were injected with 2 sgRNAs targeting the mouse Fpr2 coding sequence. The injection mixture also included a dsDNA plasmid template containing homology arms flanking the human FPR2 (ALX) coding sequence. The mouse Fpr2 coding sequence was replaced with the human FPR2 (ALX) coding sequence via CRISPR-mediated homology-directed-repair. Founders were identified by PCR across each homology arm as well and sequence-verification across the entire knock-in allele. Founders were backcrossed to C57BL/6J mates for at least 5 generations.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
