This sequence was obtained by 5'RACE PCR from a 129P2/OlaHsd mouse embryonic stem (ES) cell line (E14TG2a.4) with an insertional mutation from gene trap vector pGT1Lxf ,   (Properly spliced).

BayGenomics and SIGTR gene trap ES cell lines have a 5 prime sequence tag, therefore you are dealing with a conventional gene trap. This means your gene trap vector is located in the intron immediately downstream transcriptionally of the last exon blast hit. Vector insertions near the 5' end of a gene are more likely to result in complete gene inactivation than insertion at the 3' end, because a greater percentage of a gene's exons are prevented from being transcribed and translated. Not all genetrap cell lines provide 100% gene knock out. Please see additional information about Gene Trap lines on our ES cell lines information page.

Genotype Determination

This mouse line was confirmed via generic b-Geo screening; we do not have any information regarding the exact genomic insertion site of the vector within the intron. We recommend blasting the sequence tag to estimate the location of a gene trap. The vast majority of gene trap lines have been correctly identified, as judged by DNA sequencing (i.e. repeat determination of the DNA sequence tag when the ES cell line is requested). However, incorrectly identified gene trap cell lines are occasionally encountered, unless further verification was done, such as 5 RACE (Rapid Amplification of cDNA Ends), to check if the sequence of the cell line matches that which is published by the donating institution. We recommend that recipients of gene trap mouse lines re-verify the identity of the ES cell line by RT-PCR and/or Southern blot analysis. Strain Development
The ES cell line was injected into C57BL/6J host blastocysts to generate chimeras.  The chimeras were then crossed with C57BL/6J mice to determine germline transmission.  The resulting heterozygous males were cryoarchived for distribution.

Colony and Husbandry Information

*During development at MMRRC at UCD, mating was done with heterozygous males; viability, fertility and other breeding statistics for heterozygous females and homozygotes are unverified, but presumably would be similar to the background strain.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
