Genetic Alterations
A total of 54 bp of the exon encoding the CH1 domain were replaced in a murine gamma 3 allele heavy chain allele with a targeting vector containing the diphtheria toxin gene and a neomycin-resistance cassette (neor) bearing a beta-actin promoter.

Phenotype
Homozygous: The homozygous gamma 3 -/- mice lack serum IgG3 (at detectable levels). They appear to be more susceptible to infection with type 3 Streptococcus pneumomoniae.

Heterozygous: The heterozygotes are more similar to the wild-type mice. They produce serum IgG3.

Strain Development
This line was created by gene targeting with a neomycin-selectable gamma 3 heavy chain gene vector originating from a lambda129SV genomic library. The targeting vector was then introduced into E14-1 ES cells. The targeted ES cells were injected into C57BL/6 blastocysts and these blastocysts were transferred to the uteri of pseudopregnant C3HB16 females. Resulting males (agouti/C57BL) expressing a chimeric coat color (.25% agouti/chinchilla agouti color) were bred to National Institutes of Health Swiss Black mates. Germline chimeras were defined as producing progeny with an agouti coat color. Male and female agouti littermates (heterozygous agouti/C57BL/Swiss) were then crossed to obtain homozygous knockout (KO) pups. Homozygous wild-type (WT) littermates were saved and bred as controls.

After some time, mice bearing the defective gamma 3 heavy chain allele were backcrossed for 10 generations to BALB/c mice. These mice were then inbred as Line 632.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
