Phenotype
Cre recombinase is expressed uniquely in adult cerebellar granule cells. Thus this line can be used for cell-type specific lesions. Homozygous mice are healthy and breed normally. However, homozygous mice will have a total knockout of the alpha6-delta subtype of GABA-A receptors which is compensated for by an increased expression of another inhibitory ion channel (Brickley et al, Nature 409, 88-92, 2001).

The mice will be best deployed as heterozygoyes, which leaves one functional copy of the alpha6 gene intact.

Strain Development
A cassette containing an HSV promoter-driven neomycin gene was used. The original Gabra6 gene targeting vector was modified in exon 8. The AflII site was changed to NotI and the NcoI site was changed to AscI. The 5 -NotI-ires-nlsCreneo- AscI-3 was ligated into the targeting vector backbone. The vector was linearized with Sal I and electroporated into the stem cell line E14.1 . Targeted clones were identified by Southern blotting of colony DNA digested with SphI and hybridizing with probe A. Since transfer to MMRRC this strain has been further backcrossed to C57BL/6J to produce the congenic strain, B6.129P2-Gabra6tm2(cre)Wwis/Mmcd (see MMRRC #15968), that is 97% C57BL/6J by micro satellite testing.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
