Genetic Alterations
A plasmid was made by cloning a 3.6kb sequence from the rat alpha1-type collagen-I promoter (Col I-3.6) upstream of the tetracycline transactivator (tTA) sequence in plasmid pUGH 15-1. A purified Col I-3.6-tTA DNA fragment was prepared and used to microinject FVB/N oocytes. Injections were carried out at the Transgenic Core Facility of the UCSF-affiliated Gladstone Foundation, according to their standard methods.

Strain Development
Founder mouse was FVB/N, and was backcrossed with wild-type FVB/N mice obtained from The Jackson Laboratory. They were maintained that way for several years, at which point the donor started mating tTA positive animals with siblings until true-breeding lines were established, and is the form in which donor maintained them at time of donation to the MMRRC.

The key difference between MMRRC:030758 and MMRRC:030768 is that MMRRC:030758 has tTA driven by a 2.3 kb Col I promoter which is active in mature osteoblasts. By contrast, MMRRC:030768 has tTA driven by a 3.6 kb Col I promoter that is known to be expressed in immature as well as mature osteoblasts. Investigators interested in restricting transgene expression to mature osteoblasts would use MMRRC:030758, whereas those interested in broad expression of the transgene in osteoblast lineage cells would use MMRRC:030768.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
