Gene Information

Rorbm1Btlr
Name: RAR-related orphan receptor beta; mutation 1, Bruce Beutler
Synonyms: 4-limb clasper
Type: Allele
Species: Mus musculus (mouse)
Chromosome: 19
Alteration at locus: Chemically Induced
Rorb
Name: RAR-related orphan receptor beta
Synonyms: RZR-beta, Nr1f2, Rorbeta, hstp
Type: Gene
Species: Mus musculus (mouse)
Chromosome: 19
Alteration at locus: Chemically Induced
NCBI: 225998
Homologene: 38250
Genetic Alterations
ENU-induced T to C transition at position 127846 of the Rorb genomic DNA sequence (Genbank genomic region NC_000085 for linear genomic sequence of Rorb). The mutation occurs in the third exon of either of the two Rorb splice variants, each of which contains 10 total exons. The mutated residue corresponds to nucleotide 864 and nucleotide 330, respectively, in the cDNA sequences of Rorb isoform 1 and isoform 2. The mutation results in a serine to proline substitution at amino acid 76 and amino acid 87, respectively, of ROR?-1 and ROR?-2.
Genotype Determination
Phenotype
Homozygous: The 4-limb clasper phenotype was identified as a visible variant among G3 mice. Homozygous 4-limb clasper mice clasp all four limbs together when held by the tail, and lift hind legs excessively high when walking. Homozygous males have not been observed to breed. The phenotype of homozygous 4-limb clasper mice closely approximates that of Rorb-/- mice, suggesting that the 4- limb clasper mutation abrogates much of the function of mutant ROR?. The expression, localization, and function of the mutant protein have not been tested.

Heterozygous: Wild type.
Mammalian Phenotype Terms
Allelic Composition: Rorbm1Btlr/Rorbm1Btlr (Genetic Background: C57BL/6J-Rorbm1Btlr )

Strain Development
Original mutant was a C57BL/6J G3 ENU- induced mutant; all subsequent crosses to maintain strain were on C57BL/6J background only.

Colony and Husbandry Information

Random intra-strain mating. Homozygous females able to breed; heterozygous males not yet tested for breeding.

Colony Surveillance Program and Current Health Reports

Mice recovered from a cryo-archive will have health surveillance performed on recipient females. Health reports will be provided prior to shipment. If you require additional health status information, please email mmrrc@ucdavis.edu .

Order Request Information

Limited quantities of breeder mice (recovered litter) are available from a cryoarchive; recovered litter usually available to ship in 3 to 4 months.

Cryopreserved material may be available upon request, please inquire to mmrrc@ucdavis.edu for more information.

Distribution of this strain requires submission of the MMRRC Conditions of Use (COU). A link to the COU web form will be provided via email after an order has been placed; the form should be completed then or the email forwarded to your institutional official for completion.

The donor or their institution limits the distribution to non-profit institutions only.

Additional charges may apply for any special requests. Shipping costs are in addition to the basic distribution/resuscitation fees. Information on shipping costs and any additional charges will be provided by the supplying MMRRC facility.

MMRRC Item # Description Distribution Fee / Unit (US $) Units Notes
031040-UCD-EMBRYO Cryo-preserved embryos $1,038.00 / Non-Profit Aliquot Approximate quantity2 : 20-40 embryos / aliquot
031040-UCD-SPERM Cryo-preserved spermatozoa $546.25 / Non-Profit Aliquot Approximate quantity3
031040-UCD-RESUS Litter recovered from cryo-archive $3,033.00 / Non-Profit Litter Recovered litter4; additional fees for any special requests.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter.
