    Homozygous: Homozygous animals develop normally during the gestation period, and by size, and shape are indistinguishable from their heterozygous and wild type litter mates. They die within half an hour after birth, unable to inflate their lungs, and breath independently.

    Heterozygous: Heterozygous animals are healthy, and productive.

    Strain Development

    The founders chimeras bred against C57BL/6 with the selection by coat color (Black-agouti). All the offsprings were analyzed by PCR and some transgenics confirmed by Southern Blot. All of the transgenic animals had Neo+ cassette in place. To excise the Neo cassette in vivo, animals bred for at least 5 generations with B6;SJL-Tg(ACTFLPe)9205Dym/J mice Jackson Stock#003800. This transgenic strain expresses a variant of the Saccharomyces cerevisiae FLP1 recombinase gene under the direction of the human ACTB promoter. It excises the DNA sequence at frt sites. The resulting offsprings were selected by PCR for complete Neo cassette removal. Neo- animals were backcrossed for at least 10 generations against C57BL/6 mice. Each generation was genotyped by PCR, and periodically confirmed by Southern Blots. The final backcrosses are maintained as 2 heterozygous lines. All the breedings are documented.

    A sample from this MMRRC strain was analyzed using the Mouse Universal Genotyping Array (MUGA) and MMRRC computational tools were used to assess the genetic background. A summary of the data can be found here.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
