Phenotype
Homozygous: Mice that are homozygous or hemizygous for the targeted mutation are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express the reverse tetracycline-controlled transactivator (rtTA) protein under the control of the human CD68 promoter. The donating investigator indicates that this promoter specifies expression to a mature stage of macrophages. When mated to a second transgenic strain carrying a gene of interest under the regulatory control of a tetracycline-responsive promoter element (tetO), expression of the gene of interest may be regulated by the tetracycline analog, doxycycline (dox). In the presence of dox, transcription of the target gene is induced in cells where rtTA is expressed. This strain provides a Tet-On tool that allows the inducible expression of genes in mature macrophages.

Strain Development
The CD68-rtTA-M2 transgene was designed with a 2.9 kb promoter sequence from the human CD68 gene followed by a 83 bp fist intron (IVIS), a reverse tetracycline-regulated transactivator (rtTA) gene, and bovine growth hormone polyA tail. This 4.5 kb transgene was microinjected into the male pronuclei of two cell (C57BL/6 x CBA) mouse zygotes. Founder line 3 (CD68-MP3) was established and backcrossed to C57BL/6 mice for at least 11 generations. The donating investigator reports that the insertion site has been mapped to Chr 1.

While maintaining a live colony, these mice are bred as hemizygotes.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
