Genotype Determination

Phenotype
Homozygous: Animals homozygous for the 373 congenic region are lean compared to the background strain. When 373 mothers foster B6By female pups, the B6By female progeny are significantly leaner as adults than B6By females raised by their biological B6By mothers (P-value =0.017, ANOVA with Bonferroni posthoc).

Heterozygous: In an F1 reciprocal cross, where 373 females were bred to B6By males and vice versa, heterozygous female progeny who inherited congenic alleles from their mother were significantly leaner than females who inherited their congenic alleles from their father (P-value = 0.002, ANOVA with Bonferroni posthoc).

Strain Development
The B6.C-Tyrc H1b Hbbd/ByJ ("B6CH1" - JAX Strain #000383) congenic and its background counterpart C57BL/6ByJ ("B6By" - JAX Strain #001139) were initially purchased from The Jackson Laboratory. The mice were maintained as a homozygous colony. The B6CH1 congenic contains a 24 centiMorgan (cM) donor region retaining the H1 histocompatibility locus from BALB/cByJ on chromosome 7 on a B6By background. The D7Mit373 subcongenic of the B6CH1 strain was developed through a traditional congenic breeding scheme. After initial intercross of B6CH1 and B6By, mice were backcrossed to B6By for at least 10 generations so that >99.9% of alleles were B6By and only the BALB/cByJ allele of D7Mit373 was retained on chromosome 7. The mice were maintained as a homozygous colony until donation to the MMRRC.

Colony and Husbandry Information

In the Donor's animal rooms, the light cycle was 14/10 hrs light/dark, the ambient temperature was 21+/-2C, and >25% humidity. All breeding pairs and their pre-wean litters were housed in shoebox cages with distilled water bottles, received autoclaved cotton nestlets, and mouse igloos as enrichment. LabDiet Chow 5015.

All non-breeding mice were housed in shoebox cages with distilled water bottles or automatic watering, received autoclaved torn pieces of egg-tray flats as enrichment, and were fed purified experimental diet or LabDiet Chow 5001. Any detrimental effects on mice NOT given the above treatments are unknown.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
