Genetic Alterations
The targeting vector deletes exons 20-24 of the gene Muc19/Smgc. Exon 20 represents the third exon utilized for Muc19 transcripts. The selection cassette PGKneolox2DTA was used and the STOP cassette, pBS302 inserted immediately downstream of the 3’ loxP site bordering the neomycin coding region. A single targeted 129SvEv ES cell clone (CMT1-1) was injected into blastocysts (C57BL/6 x DBA).

Phenotype

Homozygous: Homozygous knockout mice display no discernable physiological defects. Mucous cells of glandular tissues expressing Muc19 and no other gel-forming mucin are smaller and more serous-like.  Mice are normal in all other aspects including salivary flow rates, protein and glycoprotein profiles of whole saliva and of glandular homogenates from each of the three major salivary glands (excluding Muc19 expression), body weights, activity levels, size and appearance of internal organs. 

Heterozygous: Cells of heterozygotes or that express a second gel-forming mucin appear as those of the wild type.

Strain Development
Five male chimeras (129S/C57B6J) were mated to Black Swiss females (NIHBS) and produced a single male F1 agouti from 19 litters. The agouti male was mated to NFS/NCr females at the University of Florida and heterozygous progeny intercrossed to generate two Muc19-/- females that were mated to a male transgenic cre-deleter B6.C-Tg(CMV-cre)1Cgn/J (Jackson Labs). Progeny were intercrossed and three resultant Muc19+/- males (neo-, cre-) used to initiate five backcrosses to NFS/N mice. N5 heterozygotes were intercrossed to establish two closed colonies, one Muc19-/- and the other Muc19+/+ controls.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
