Genetic Alterations
Knockin mice have EGFP inserted in-frame within exon 21 of the gene Muc19/Smgc. Exon 21 represents the fourth exon utilized for Muc19 transcripts. As a result, chimeric transcripts encode only the first 69 residues of Muc19, including the signal peptide that directs translation products to the secretory pathway.

Phenotype

Homozygous: Homozygous knockin mice display no discernable physiological defects. Mucous cells of glandular and mucosal tissues expressing Muc19 and no other gel-forming mucin are smaller and more serous-like with EGFP-positive exocrine granules.  Mice are normal in all other aspects including salivary flow rates, protein and glycoprotein profiles of whole saliva and of glandular homogenates from each of the three major salivary glands (excluding Muc19 expression and expression of EGFP), body weights, activity levels, size and appearance of internal organs. 

Heterozygous: Cells of heterozygotes or of those that express a second gel-forming mucin are similar in ultrastructure to wild-type mucous cells except for EGFP-expressing secretion granules.

Strain Development
A single male chimera (129S/C57B6J) produced from a 129S6 ES cell clone (CMT1-1 cells) was mated to Black Swiss females (NIHBS) and produced 31 heterozygous F1 agouti mice. Four breeder pairs of heterozygous F1 agouti mice were used to establish a closed colony of Muc19-EGFP knockin mice.


Three ES cell clones were expanded and injected into C57Bl/6 blastocysts, resulting in a total of 32 chimeric mice. Chimeric males were mated to Black Swiss (NIHBS) females. One male breeder produced 31 agouti heterozygous F1 progeny, from which four breeder pairs were established at the University of Florida and interbred to generate wild-type and heterozygous animals for EGFP expression experiments.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
