Genetic Alterations
Using GENSAT BAC engineering system, the Cre recombinase gene was introduced upstream of the ATG start codon of the gene of interest on a selected BAC clone. The inserted gene has its own ATG start codon and polyadenylation signal. This modification results in the expression of Cre recombinase that is under the control of the promoter and regulatory elements of the gene of interest that are present on the BAC clone. The BAC address for this line is RP23-354N13. SNP analysis has shown the transgenic insertion to be on chromosome 16 between SNPs rs4152638 and rs4158907.
This congenic strain carries the same mutation as MMRRC:032081 and was originally characterized on a STOCK background before backcrossing to C57BL/6J for at least 8 generations. It should be noted that the phenotype could vary from that originally described. Strain Development
The original transgenic mice were created in the GENSAT project in collaboration with the Intramural Program of the National Institute of Mental Health. The transgenic mice have a modified BAC-cre injected into pronuclei of FVB/N fertilized oocytes. Offspring of founders were crossed to a Soriano Rosa26/stop/EGFP reporter line. The MMRRC backcrossed this transgene from the mixed background to C57BL/6J, selected for mice with only the cre expression, and eliminated the Rosa26-GFP from the line. Mice were determined to be congenic on C57BL/6J by SNP analysis.

Colony and Husbandry Information

In general, GENSAT BAC transgenic mouse lines should be utilized in the hemizygous state. Breeding of the BAC transgene to a homozygous state carries an increased risk of generating an unwanted phenotype for two reasons. First, the transgene inserts at random into the genome. Breeding the transgene to homozygosity results in mice that carry this insertion on both chromosomes, increasing the probability that the insertion will result in an observable phenotype. Second, BAC transgenes may carry passenger genes that are also expressed from the BAC vector. Breeding to homozygosity doubles the number of these genes carried in the transgenic mouse, increasing the possibility that a phenotype will result as a consequence of increased gene dosage. In our experience, observable phenotypes are rare in BAC transgenic mice when carried in the hemizygous state.

1 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

3 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.
