Genetic Alterations
The targeting construct for creating the oxed Aqp11 gene, with loxP sites anking exons 2 and 3, was generated by insertion of PCR ampli ed genomic DNA segments spanning over the Aqp11 gene sequence into a modi ed pKOScrambler(FRT-loxP) vector. The loxP site was inserted upstream of exon 2 on the Aqp11 gene. An FRT-flanked neo cassette with a 5' loxP site was inserted downstream of exon 3. The neomycin phosphotransferase expression cassette was deleted from the floxed Aqp11 allele by transient transfection in the targeted ES cells with a Flp-recombinase expression plasmid leaving exons 2 and 3 floxed. The Aqp11 floxed mice were bred with B6.Cg-Tg(Alb-cre)21Mgn/J to remove the floxed exons.

Phenotype

Homozygous: No observable phenotype.

Heterozygous: No observable phenotype.

Expected Phenotype After Cre-excision: In the liver: the mice show vacuolation of the cytoplasm in periportal hepatocytes, after metabolic challenge: for example fasting/re-feeding, or administration of high doses of free amino acids. The vacuoles are derived from enlarged RER-lumen. The changes in the liver disappear after 24 hours. In the kidney: the mice show vacuolation of the cytoplasm in proximal tubule cells in the early neonatal period, and tubular damage. From 2 weeks of age, the mice show gradual increase in tubule diameter, resulting in development of polycystic phenotype by 3-4 weeks of age. The kidney phenotype is lethal in most animals.

Strain Development
The targeting construct was linearized and electroporated into CJ7 cells derived from 129S1/Sv-Oca2+ Tyr+ Kitl+ mice. Targeted ES cells were injected into B6D2F2 mouse blastocysts. Chimeric males were bred with C57BL/6 females, with screening for germ-line transmission by identification of agouti offspring. For cre-excision, Aqp11 flox mice were bred with B6.Cg-Tg(Alb-cre)21Mgn/J to generate the liver specific KO.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
