Genetic Alterations
In the transgenic portion of this strain, a human promoter drives ubiquitous expression of the mouse long form of the prolactin receptor. The targeted mutation consists of a neomycin cassette inserted into Prlr exon 5, creating an in-frame stop codon.

Strain Description

Phenotype

Homozygous: Female mice expressing only the long form of the prolactin receptor are infertile. The corpus luteum in the ovary does not form properly due to impaired vascularization and luteinization and progesterone production is abnormal and therefore implantation of the embryos is impaired. Male mice expressing only the long form of the prolactin receptor are fertile, but appear to have a metabolic defect where they become obese and potentially insulin resistant.

Hemizygous: The hemizygous mice who express the transgene will express both the long and short forms of the prolactin receptor.

Strain Development
To create mice with ubiquitous expression of PRLR-L, a targeting construct was generated. The Prlr-L cDNA was generated by PCR and subsequently subcloned into the EF1A-pPolyIII vector. This targeting vector (EF1-RL), used to generate mice with ubiquitous expression of PRLR-L, contains EF1A promoter, SR enhancer region, long isoform of the prolactin receptor cDNA, and a portion of human growth hormone (hGH) cDNA, a tag to identify transgenic mice. Following confirmation of activity of the transgenic vector, the constructs were linearized and injected into pronuclei of fertilized oocytes from FVBN mice. Transgenic mice were identified by PCR and Southern blotting using primers/probes to the hGH tag. Transgenic mice were backcrossed with prolactin receptor null mice to generate mice expressing only the long form of the prolactin receptor in a ubiquitous manner.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
