Genetic Alterations
This line is an Aff4 knockout, swapping exon 2 in-frame with a lacZ reporter.

More specifically, an isogenic targeting vector was designed to delete Aff4 exon 2 (also known as Af5q31), which contains the start codon. To this end, a 681-bp HincII fragment containing all of exon 2 and flanking sequences was replaced with an E. coli lacZ and neomycin resistance (neo) cassette. This is predicted to delete the original starting ATG as well as the first 54 amino acids, even if translation were to start at the next in-frame ATG after exon 2. These 54 amino acids contribute to the highly conserved NHD domain of the AF4/FMR2 gene family. The design also provided an in-frame replacement of exon 2 with lacZ, to allow in vivo tracking of Aff4 expression. The left arm of the construct is 6.8 kb, composed of a 3.9-kb HindIII fragment and a 2.9-kb fragment from HincII to HindIII. The right arm is 5.8 kb, consisting of 5.8-kb PstI/XbaI fragment and a 100-bp PstI/HincII fragment.

Strain Development

The targeting vector was introduced into AB2.2 (129S7/SvEvBrd) embryonic stem cells and G418-resistant colonies were selected. Southern blot hybridization confirmed correct integration in two clones, which were expanded and injected into C57BL/6 blastocysts. Resulting chimeras were backcrossed to C57BL/6 for several generations. Heterozygotes from N2 through N5 were sib-mated to generate homozygous mice. Southern blotting and PCR were used for genotyping, and complete absence of Aff4 mRNA containing exon 2 was confirmed by northern blotting, using an exon 2 specific probe.

Note: Aff4 mRNA was detected using other probes, suggesting that there is a truncated mRNA lacking exon 2, even though no protein could be detected using Af5q31 polyclonal antibodies.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
