Genetic Alterations
The Il13a1 locus which is located on the X chromosome was disrupted by homologous recombination between the endogenous Il13a1 wild-type allele of male karyotyped BALB/c embryonic stem cells with the Il13a1 targeting vector. Confirmation of homologous recombination at the targeted allele was demonstrated by Southern blot. Following confirmation of the disrupted allele, chimeric blastocysts were generated by micro injection into developing blastocysts derived from C57BL/6 female donors. Males from this litter displaying the highest degree of white coat color were bred to BALB/c females and progeny were genotyped by PCR to assess for germline transmission of the disrupted Il13a1 allele. Only females having white coats and displaying the germ line transmission by PCR were bred back to a wild-type BALB/c male to generate Il13a1-/- hemizygous males, which were than bred back with heterozygous females to generate Il13a1-/- homozygous females. To confirm deletion of exons 7,8,9 in transcripts from the disrupted Il13a1 allele, mRNA was extracted from intestinal epithelial cells of Il13a1-/- and Il13a1+/+ littermate controls, since intestinal epithelial cells express IL-13R alpha1 in non-manipulated mice. cDNA was synthesized from mRNA by reverse transcription PCR. The bands were purified and sequenced to confirm deletion of exons 7-9 in the Il13a1-/- mice. Speed congenics was used to move the Foxp3-GFP from the C57BL/6 to the SJL strain

Strain Development
The SJL, SJL/IL-13R alpha1 -/- and the SJL FoxP3-GFP mice were used to develop this line. SJL/IL-13R alpha1 -/- was bred to the SJL FoxP3-GFP to produce heterozygous female SJL/ IL-13R alpha1-/- FoxP3-GFP. These females were bred back to a SJL male in order to get a hemizygous male that has both genes on the X chromosome. This male then was bred back to SJL females to produce the heterozygous females and hemizygous males. The heterozygous females were bred to the hemizygous male to produce both homozygous females and hemizygous males. These were then used to continue the line. All steps were confirmed by PCR on their on their homozygous or heterozygous status.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
