Genetic Alterations

Homozygous synapsin I, II and III triple knockout mice exhibit seizures, behavioral defects and altered basal transmission at excitatory and inhibitory synapses. These mice may be useful in studying the role of synaptic transmission.

Phenotype
Syn1, Syn2, and Syn3 are members of the synapsin family of neuronal phosphoproteins and localize to the membranes of synaptic vesicles. Synapsin proteins are associated with autism. The synapsin I, II, and III knockout alleles all have a neo cassette replacing exon 1 of their respective genes. Homozygous triple knockout (TKO) mice exhibit a decrease in the rate of synaptic depression during trains of stimuli and a reduction in the number of vesicles in reserve pool of excitatory synapses. In contrast, basal transmission at inhibitory synapses is reduced, but kinetics are unaffected and the total number of synapses is reduced. Behavioral phenotypes include delayed whisker, eye and ear reflexes, abnormalities in coordination and balance, especially in the hindpaws, and deficits in spatial learning and memory. Beginning at two months of age, mice experience mild and transient spontaneous seizures. Homozygous mice are viable and fertile with no reported reproductive deficiency.

Strain Development
The targeting vectors for all three alleles were designed by Dr. Paul Greengard (Rockefeller University). In all cases (Syn1-Chr X, Syn2-Chr 6, and Syn3-Chr 10), a PGK-neomycin cassette replaced exon 1. The constructs were electroporated into 129-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts, and chimeric males were bred with C57BL/6J females to establish the three colonies. Syn2tm1Pggd(MMRRC 041437)-deficient mice, Syn3tm1Pggd (MMRRC 041435)-deficient mice, and Syn1tm1Pggd (MMRRC 041436)-deficient mice were combined to generate a homozygous triple knockout.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
