Genetic Alterations
These inducible NCX1 transgenic mice express rat Slc8a1 (or NCX1) under the direction of a cardiomyocyte-specific promoter (Myh6) fused with tetO. Following DOX induction, bitransgenic mice exhibit a 42% increase in SLC8A1 current amplitude and prolonged action potential duration. This strain may be useful for studying heart disease.

These inducible NCX1 transgenic mice express rat Slc8a1 (solute carrier family 8 (sodium/calcium exchanger), member 1 or NCX1) under the direction of a modified mouse alpha-myosin heavy chain minimal promoter (Myh6) fused with a tetracycline operator promoter (tetO). SLC8A1 is a Na+/Ca2+-exchanger that mediates Ca2+ efflux and influx during the excitation-contraction cycle in cardiac muscle. Mice hemizygous for the transgenic insert are viable and fertile. It is not known if homozygotes are viable. When mated to mice carrying MHC-tTA (also known as Tg(Myh6-tTA)55Rbns), SLC8A1 expression is directed to cardiac myocytes and regulated with tetracycline or its analog doxycycline (DOX). Following DOX induction, bitransgenic mice exhibit a 42% increase in SLC8A1 current amplitude and action potential duration is prolonged. This strain may be useful for studying heart disease.

Strain Development
The inducible NCX1 transgene contains the rat Slc8a1 gene driven by a modified mouse alpha-myosin heavy chain minimal promoter (Myh6) fused with tetracycline responsive element (TRE or tetO). The transgene was injected into fertilized FVB/N mouse eggs. Founder B, carrying 11 copies of the transgene, was subsequently established and bred to FVB/NJ for several generations. Upon arrival at The Jackson Laboratory Repository, the mice were bred with FVB/NJ mice for at least one generation to establish the colony.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
