Genetic Alterations
A targeting vector was designed for the insertion of a complex cassette into the 5′-region of the c-Kit gene. Lox66 and lox71 sites in a head-to-head orientation flank splice acceptor (SA): an inserted internal ribosomal entry site (IRES):eGfp as well as two polyadenylation/stop signals. Prior to Cre recombination, the c-Kit lox66–71 allele will transcribe a full-length c-Kit transcript. Upon activation of Cre, the inserted genes are inverted by the flanking lox66 and lox71. The IRES downstream from the c-Kit exon 8 allows for a bicistronic expression of eGfp and for an activation of stop signals from two polyadenylations, thereby creating a transcription stop of the downstream c-Kit gene

Strain Development

The targeting vector was linearized by AhdI and then transfected by electroporation into C2J ES cells derived from C57BL/6-Tyrc-2J/J mice. Colonies doubly resistant for G418 and gancyclovir were screened by Southern blot analysis for homologous recombination with XbaI digestion using 3′-flanking probe. Subsequently, the Neo was excised from the targeted allele in the homologous recombinant ES clones by transient expression of Flippase-puromycin plasmid. After screening of Neo-excised ES clones by Southern blot hybridization with BglII digestion using 3′-flanking probe, the screened ES clones were used for microinjections with C57BL/6 blastocysts. Generated chimeric mice were bred with C57BL/6-Tyrc-2J/J female mice, and their albino-looking offspring were genotyped to confirm germline transmission of the c-Kit lox66–71 allele. The strain was then bred to non-albino C57BL/6 mice  to remove the Tyrc2J allele.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
