Genetic Alterations
Apoe App double knock-out mice exhibit several of the same characteristics as mice carrying the individual KO alleles. In double KO mice on a high fat diet, lipid accumulation is attenuated and collagen and muscle content is increased. This strain may be useful for studying the role of APP in plaque formation.

Phenotype
Mice homozygous for the apoliprotein E (Apoe) and amyloid beta precursor (App) knockout mutations exhibit traits observed in single KO mice such as: an increase in total plasma cholesterol levels, fatty streaks in the proximal aorta progressing to pre-atherosclerotic lesions, decreased weight, decreased motor activity and forelimb grip strength, and reactive gliosis. On a high fat diet, double KO mice exhibit an attenuated lipid accumulation and increased collagen and muscle content. This strain may be useful for studying the role of APP in plaque formation.

Strain Development
A targeting vector containing neomycin resistance cassette was used to disrupt part of exon 3 and part of intron 3 of Apoe gene. The construct was electroporated into 129P2/OlaHsd-derived E14TG2a embryonic stem (ES) cells. The resulting chimeric animals were backcrossed to C57BL/6J for 10 generations.

A targeting vector containing neomycin resistance and herpes simplex virus thymidine kinase genes was used to disrupt a region of the App gene encoding the promoter and exon 1. The construct was electroporated into 129S7/SvEvBrd-derived AB2.1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. The resulting chimeric animals were backcrossed to C57BL/6J for 10 generations.

Mice homozygous for the Apoetm1Unc mutation were intercrossed with mice homozygous for the Apptm1Dbo; subsequent intercrosses generated mice homozygous for both mutations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
