Genetic Alterations
The human myelin basic protein gene was truncated in the cDNA construct to code for amino acids 1 to 64 only (MBP). The MBP cDNA fragment was subcloned in-frame with EGFP cDNA. The protein is specifically neuronally derived because of the murine Thy1 promoter.
Phenotype

Homozygous: The transgenic MBP1-EGFP shows no obvious phenotype, however crossing it with an Alzheimer's disease mouse model improved behavior and reduced amyloid accumulation.

Hemizygous: Normal.

Strain Development
Transgene plasmid was sent to the UC Irvine Transgenic Mouse Facility. The Thy1-MPB1-EGFP transgene was cleaved out of the plasmid with EcoRI and PvuI and pronuclear injected into C57Bl/6NTac egg donor. Pronuclear injections produced 3 positive founders. All the founders were breed to C57Bl6/NTac wild-type mice. Founder B did not produce pups. Male founder A pup had a very small amount of neuronally derived expression of MBP and GFP. Female founder C pups were kept, because of the MBP and GFP expression in the brain. Heterozygote pups were bred to C57Bl/6NTac to produce a few litters of heterozygote pups; null wild-types were eliminated. Het males and females were bred to produce homozygotes; the resulting males were crossed to C57Bl/6NTac wild-type mice to check for true homozygosity.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
