Phenotype

Homozygous: The mice produce no detectable Prdx6 mRNA, protein or enzyme activities. These mice accumulate phospholipids in the lung over time and are more sensitive to oxidative stress than wild-type mice.

Hetero/Hemizygous: Heterozygous mice were found to produce around 50% of the levels of Prdx6 mRNA and protein as the homozygous null mice.

Strain Development
The targeting vector contained both the Herpes TK gene and a neomycin resistance cassette. The targeting vector was linearized and transfected into the 129/Sv ES cell line, R1. The transfected ES cells were selected in medium containing 300 μg/ml of G418 and 2 μM ganciclovir. Colonies resistant to both drugs were screened by Southern blot analysis using a 0.2 kb EcoRI genomic fragment which is external to the genomic sequence in the targeting vector. Using this probe, the signal from the endogenous mouse 1-cysPrx gene was about 6.4 kb and the signal from the targeted allele was about 8.2 kb. Clones containing the targeted allele were microinjected into C57BL/6J blastocysts and embryos were re-implanted into the uterine horns of foster mothers. Male chimeras with more than 95% agouti coat color were chosen to breed with C57BL/6J female mice (Taconic, Germantown, NY, USA). Mice were tested for germline transmission and those offspring containing an inactivated copy of the 1-cysPrx gene were backcrossed to >99.9% C57BL/6J, based on genotyping performed by Jackson Laboratories using microsatellite markers. These mice were then bred to each other to produce homozygous Prdx6 null mice.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
