Genetic Alterations
A 13kb Pitx1 enhancer 132kb 3' of the promoter, shown to topologically interact with the target gene through 3C and 4C, was deleted via CRISPR. The deleted sequence spans chr13:56,055,928-56,068,947.

Phenotype

Homozygous: The mice no longer topologically interact with the enhancer region. At E11.5 Pitx1 expression is reduced by 15% in the hindlimb and 39% in the mandibular arch. Their morphology is overtly normal and exhibit normal viability.

Hetero/Hemizygous: The heterozygous mice display a normal phenotype. In depth molecular analysis was not conducted on the hets.

Cre-excised Phenotype: Undetermined. 

Strain Development
A conditional allele was produced by using CRISPR to concurrently flox the 13kb enhancer sequence (the Pitx1 distal enhacner, or PDE) through pronuclear injection by the Yale Transgenic Facility. The founder mouse containing the two loxp sites was bred back to wt B6 mouse. This mouse was crossed with an actin cre expressing mouse to produce PDE-/- mice. These mice were then used in sib crosses to expand the line.
