Genetic Alterations
Exon1(alpha) was replaced with a translation start site followed by tdTomato cDNA, an SV40 polyadenylation signal, and a FRT-flanked neo cassette. The neo cassette was retained, as it proved to enhance tdTomato expression. It may be removed via Flp-mediated recombination.

    Homozygous: Mostly normal phenotype. Mice have cancer-prone phenotype with aging, causing reduced lifespan, as null for p16INK4a expression so similar to p16 KO (PMID:11544531). Express tdTomato. No known embryonic lethality.

    Hetero/Hemizygous: Normal phenotype. Express tdTomato. No known embryonic lethality.

    Strain GQC Summary
    Incipient congenic strain carrying the tm4Nesh allele of the Cdkn2a gene on C57BL/6 and 129 mixed background. The allele of interest present on chromosome 4 is on the donor background (129S4/SvJaeJ). The donor background is also present in other chromosomes. At least one additional unexplained genetic background was detected. 129S4/SvJaeJ, 129S6/SvEvTac, C57BL/6J, and C57BL/6NTac were detected with diagnostic alleles. The genome of this strain is not fully replicable because up to 11.2% of the genome is segregating for the donor background, at least 60.9% of the genome is segregating for the substrains of the primary background, and at least 1.1% of the genome is segregating for the unexplained background. This strain carries a potentially active allele of red (or similar) fluorescent protein with unknown zygosity. This GQC report is based on the MiniMUGA G3 2024 pipeline using 2 samples genotyped in 2019.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered.     2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
