A floxed neo STOP cassette is inserted in the Hand1 5'UTR along with two point mutations:
Cre recombination removes the STOP cassette and allows for endogenous level of expression in tissues where Cre and Hand1 expression overlap.
Homozygous: lethal at E9.5 it is a conditional actionable allele
Hetero/Hemizygous: heterozygous mice are viable and fertile when there is no cre recombination
Cre-excised Phenotype: undetermined
Generation of targeting vector, which introduces a floxed neomycin stop cassette into the 5’UTR of Hand1 along with point mutations to produce p.Thr107Ala/p.Ser109Ala (this strain) or p.Thr107Asp/p.Thr109Asp (MMRRC:43548). Electroporation of CCE916 ES cells followed by G418 selection. Clonal expansion followed by Southern blot RFLP analysis (or PCR) to confirm recombinant clones. Injection of desired clones into C57BL/6 blastocysts to produce chimeras which were then mated to C57BL/6. The strain was maintained on a mixed C57BL/6, 129/SvJ, NIH Swiss background
