Germline knock-out App mice have a deletion of exon 3 of the amyloid beta (A4) precursor protein gene (App) - this eliminates APP protein expression in the cortex. These mice may be useful for studying APP function and Alzheimer's disease.

Amyloid beta (A4) precursor protein (APP) is a type I integral membrane protein that is cleaved to form a number of peptides, in particular, the amyloid-β-peptide, a component of the amyloid plaques associated with Alzheimer's disease.

The germline knock-out App allele has exon 3 deleted. APP protein expression is eliminated in the cortex, however, cortical layer markers, astrocyte markers and microglial markers are not disrupted. Homozygous mice are viable and fertile. No gross morphological differences are observed between embryonic wild-type and knock-out brains. These mice may be useful for studying APP function.

Strain Development
The targeting vector was designed to insert a loxP site in intron 2 and a frt-flanked neomycin selection cassette in intron 3 followed by a second loxP site. The construct was electroporated into 129S4/SvJae-derived J1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Chimeric animals were crossed to C57BL/6 mice, and offspring were crossed to B6;SJL-Tg(ACTFLPe)9205Dym/J mice, and then crossed to BALB/c-Tg(CMV-cre)1Cgn/J to remove exon 3. Resulting mice with the germline knock-out App allele were selected and then backcrossed to C57BL/6J mice for at least five generations (and the FLPe- and Cre-expressing transgenes were removed). Upon arrival at The Jackson Laboratory Repository, mice were bred to C57BL/6J for at least one generation to establish the colony.
