These mice are a positive control for ROSAnZ∆4 (MMRRC:44026), which have the mutated lacZ gene. They have a wild-type lacZ gene at the same locus as ROSAnZ∆4.

Strain Development
Mouse strain C57BL/6-derived embryonic stem cells were electroporated with 10–20 mg of plasmids pROSAnZ or pROSAnZ∆4, linearized with SacII to generate transgenic mice. Transfected cells were selected in medium containing 0.3 mg/ml G418, and targeted clones were identified by Southern blot analysis. Several targeted clones were injected into BALB/c-derived blastocysts and implanted in pseudopregnant females (strain CD1). Chimeric mice were bred to C57BL/6J females and litters screened by coat color. Mice with black coats were checked by Southern blot analysis for the presence and structure of the targeted locus and bred to littermates to establish homozygosity for the targeted loci. Both ROSAnZ (MMRRC:44049) and ROSAnZ∆4 strains were maintained as homozygous breeding pairs that appeared healthy.

The ROSAnZ mouse is the control strain for the ROSAnZ∆4 (MMRRC:44026) strain, which is a useful model to demonstrate successful gene-therapy by gene-targeting in vivo. It can be used with any gene-editing method; CRISPR, lentiviral, foamy virus, rAAV, etc., by restoring the function of the deleted lacZ gene detecting the color in targeted tissues. The plasmid can be recovered from target tissues and grown up in bacteria.
