Genetic Alterations
This strain carries a transgene expressing the full-length CD45 protein with a mutation of the catalytic cysteine of the phosphatase domain (p.Cys817Ser or C817S) that renders the protein without tyrosine phosphatase activity. The details of the transgene mutation are as follows:

  • nucleic acid level: c.2525G>C or c.2524T>A (based on the simplest possible exchange)

Note: The literature has been referring to this variant as "C817S" dating back to 1992 (e.g., Johnson et al., JBC 1992 - PMID:1314815) or earlier. However, the 817 numbering does not include a leader sequence that is removed from in the mature protein.

This CD45 minigene strain is one of a series of transgenic strains prepared with the CD45 minigene. The minigene was altered with a single base change to mutate the catalytic cysteine of the phosphatase domain (C817S), which renders the CD45 protein enzymatically inactive. This strain has proven useful for studies of non-enzymatic functions of the CD45 molecule on lymphocyte activation and maturation. This strain should be also suitable for similar activation and maturation studies of other hematopoietic lineages which express CD45. The strain was also instrumental in establishing the requirement for CD45 phosphatase activity for the protection of mice with reduced CD45 levels from lethal effects of multiple pathogens.

Strain Development
The CD45-minigene transgenic construct 'VGI-0020CS', which encodes a point mutation in VGI-0020 (also known as LFAiT200 in reference PMID:12393487) was injected into C57BL/6 zygotes. One founder "CSV10" was identified, which was mated to C57BL/6J mice and progeny were screened for the presence of the transgene. Mice homozygous for the transgene locus were obtained and then sibling-mated for at least 25 generations (F25+) to date.

This CD45-minigene transgenic strain on the CD45 knockout C57BL/6 background (JAX Strain #002725) expresses a full-length CD45 protein with no protein tyrosine phosphatase activity due to a mutation of the catalytic cysteine. It has the same severe immunodeficiency phenotype as the CD45 knockout strain, including T and B cell functional and maturational abnormalities. The strain also has the below average breeding characteristics of the CD45 knockout strain. Normal hematopoietic lineage and maturation-specific isoform regulation of the transgene product is maintained due to the inclusion of the natural introns between exons 3 and 9 in the transgene DNA sequence. The transgene encodes the CD45.1 allelic sequence which is detected by anti-CD45.1 antibodies. Due to the inconsistent breeding property of this strain on the CD45 knockout background, the strain is routinely maintained on the wild-type C57BL/6 background and bred onto the knockout background (JAX Strain #002725) for experimental purposes.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
