CD45-minigene transgenic strain LP8 on the CD45 knockout C57BL/6 background (JAX Strain #002725) expresses 63% of the wild-type level of CD45. This level is the same as mice with one functional copy of the wild-type endogenous allele (mice heterozygous for the knockout allele) and also the same as expressed by CD45-minigene transgenic F (MMRRC:46054). The transgene encodes the CD45.1 allelic sequence which is detected by anti-CD45.1 antibodies. This strain, like all of the CD45-minigene transgenics, expresses normal cell lineage-specific isoform regulation. The strain has no breeding, lifespan or general health deficiencies.

Strain Development
The CD45-minigene transgenic construct 'VGI-1084' was injected into C57BL/6J zygotes, and 7 founders were identified: LP4, LP5H, LP5L, LP8, LP10, LP26, and LP28. VGI-1084 contains the same CD45 minigene construct as VGI-0020 (also known as pLFAiT200 in PMID:12393487) but has a LoxP site added at each end of the minigene. The LoxP sites allow the minigene to be deleted from the genome in the presence of cre. CD45-minigene transgenic LP8 (the ‘LP’ series designating the presence of the LoxP sites) was one of the founders. The LP8 founder was mated to C57BL/6J mice and progeny were screened for the presence of the transgene. The colony was then crossed onto the CD45-null background by matings with CD45 knockout mice also on the C57BL/6 background (JAX Strain #002725). LP8 mice homozygous for the transgene locus and on the CD45 knockout background were then sibling mated for at least 10 generations to date. The LP8 mice express functional CD45 protein solely from the CD45 minigene.

CD45-minigene transgenic LP8 is one of a series of transgenic strains prepared with the CD45 minigene. Strain LP8 expresses 63% of the wild-type level of CD45, which is equivalent to the level expressed by mice with one functional copy of the wild-type endogenous allele and by CD45-minigene transgenic F. Both the heterozygous CD45 knockout and transgenic strain F mice were instrumental for establishing that CD45 reduction provides protection from lethal effects of multiple pathogens, as these strains are resistant to lethal doses of B. anthracis, Ebola, and other pathogens (published), unlike wild-type mice and strains expressing low levels of CD45. CD45-minigene transgenic strain LP8 offers a substantial benefit by having the added feature of the minigene being flanked by LoxP sequences, such that crosses of LP8 mice with cre-expressing strains result in transgene excision and 0% CD45 expression in cells expressing cre. Crosses of strain LP8 with vav-icre mice (JAX Strain #008610) produce mice with 0% expression of CD45 in hematopoietic cells in which vav is expressed. Strain LP8 is therefore suitable for generating conditional mutants lacking CD45 expression in specific hematopoietic subpopulations using the many strains available with cell type-specific cre expression. This strain should be particularly suitable for studies on the cell types responsible for the disease protection reported in mice with reduced CD45 expression. Strain LP8 is also a replacement for transgenic strain F (MMRRC:46054) for studies of the effect of CD45 expression level, and thus CD45 phosphatase activity, on lymphocyte activation and maturation. This strain should be also suitable for similar activation and maturation studies of other hematopoietic lineages which express CD45, especially using the LoxP feature with conditional cre expression.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
