CD45-minigene transgenic strain LP5H on the CD45 knockout C57BL/6 background (JAX Strain #002725) expresses 77% of the wild-type level of CD45. This strain is one of a second series of transgenic strains prepared using the CD45 minigene (PMID:12393487, PMDI:19269962, PMID:20346773) with the added characteristic of LoxP sequences flanking the minigene. The second series of transgenics, the LoxP or LP series, allows excision of the transgene locus by cre expression. The transgene encodes the CD45.1 allelic sequence which is detected by anti-CD45.1 antibodies. This strain, like all of the CD45-minigene transgenics, expresses normal cell lineage-specific isoform regulation. The strain has no breeding, lifespan or general health deficiencies.

Strain Development
The CD45-minigene transgenic construct 'VGI-1084' was injected into C57BL/6J zygotes, and 7 founders were identified: LP4, LP5H, LP5L, LP8, LP10, LP26 and LP28. VGI-1084 contains the same CD45 minigene construct as VGI-0020 (also known as pLFAiT200 in reference PMID:12393487) but has a LoxP site added at each end of the minigene. The LoxP sites allow the minigene to be deleted from the genome in the presence of cre. CD45-minigene transgenic LP5H (the ‘LP’ series designating the presence of the LoxP sites) was one of the founders. The LP5H founder was mated to C57BL/6J mice and progeny were screened for the presence of the transgene. The colony was then crossed onto the CD45-null background by matings with CD45 knockout mice also on the C57BL/6 background (JAX Strain #002725). LP5H mice homozygous for the transgene locus and on the CD45 knockout background were then sibling mated for at least 10 generations to date. The LP5H mice express functional CD45 protein solely from the CD45 minigene.

CD45-minigene transgenic LP5H is one of a second series of transgenic strains prepared with the CD45 minigene, differing from the original series by the inclusion of flanking LoxP sequences. Strain LP5H expresses 77% of the wild-type level of CD45 and fills an important gap in the sequence of CD45 expression levels for studies of effects of CD45 levels, and thus CD45 protein tyrosine phosphatase levels, on the protection from lethal pathogens, including B. anthracis, Ebola and others, afforded by reduced CD45 expression (PMID:19269962, PMID:19683682) and on lymphocyte activation and maturation (PMID:20346773, PMID:23135465). Whereas these published studies utilized the initial series of CD45 minigene transgenics, the LoxP flanked transgenic series should provide superior flexibility by providing cre-excision capability as well as a more complete spectrum of CD45 expression levels. CD45-minigene transgenic LP5H offers the substantial benefit of flanking LoxP sequences, such that crosses of the LP5H strain with cre-expressing strains result in transgene excision and 0% CD45 expression in cells expressing cre. Crosses of LP5H with vav-icre mice (JAX Strain #008610) produce mice with 0% expression of CD45 in hematopoietic cells in which vav is expressed. Strain LP5H is therefore suitable for generating conditional mutants lacking CD45 expression in specific hematopoietic subpopulations using the many strains available with cell type-specific cre expresssion. Strain LP5H, along with the other LP series of transgenic strains, is well suited for further investigations of lymphocyte activation and maturation. Importantly, this strain should also be well suited for similar activation and maturation studies of other hematopoietic lineages which express CD45, especially using the LoxP feature with conditional cre expression.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
