Genetic Alterations
The Adamts18-/- mouse (VG 12442) was generated by the trans-NIH Knock-Out Mouse Project (KOMP) and obtained from the KOMP Repository (www.komp.org). Adamts16 was deleted from these mice through CRISPR/Cas9 technology to generate Adamts16-/-;Adamts18-/-. The deletion was confirmed by Sanger sequencing (deletion of 33 bp within Exon 1, including the ATG site, see Fig. 6, PMID:31242448).

    Phenotype
    Adamts18 encodes a secreted metalloprotease restricted to branch-tip progenitor pools directing the morphogenesis of multiple mammalian organs. Adamts18 mutants displayed a weakly penetrant phenotype: duplicated ureteric outgrowths forming enlarged, bi-lobed kidneys with an increased nephron endowment. In contrast, Adamts18 mutants showed a fully penetrant lung phenotype: epithelial growth was markedly reduced and early secondary branching scaled to the reduced length of the primary airways. Adamts18 is closely related to Adamts16, in the kidney but not the lung, broad epithelial Adamts16 expression overlaps Adamts18 in branch tips.

    The Adamts18-/- mouse (VG 12442) was obtained from the KOMP Repository (www.komp.org) and deleted expression of Adamts16 (deletion of 33bp within Exon 1, including the ATG site) in these mice through CRISPR/Cas9 technology to generate the double KO. The deletion was confirmed by Sanger sequencing. Compound Adamts16/18 mutants displayed a comparable low penetrance duplicated ureteric phenotype, ruling out a possible role for Adamts16 as a functional modifier of the Adamts18 kidney phenotype. Given the predicted action of secreted Adamts18 metalloprotease, and broad expression of Adamts18 in branching organ systems, these findings suggest distinct requirements for matrix modeling in the morphogenesis of epithelial networks.
    Strain Development
    Zygotes from C57BL/6NTac-Adamts18tm1.1(KOMP)Vlcg mice (KOMP repository; Project ID VG12442) were bred to C57BL/6N initially to expand the line for about 3 generations. The heterozygous Adamts18 mice were intercrossed to generate a homozygous line and maintained by sib-mating for about 5 generations. The Adamts16 gene was knocked out by CRISPR/Cas9 in the Adamts18 line and the resulting mice were intercrossed for about 6-8 generations resulting in a double knockout of the non-segregating Adamts16 and Adamts18 alleles.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients.
