Genetic Alterations
The Chd5 gene was constitutionally removed exons at 12 and 13, changing the reading frame leading to a stop codon, resulting in no detectable protein. The mice have no Neo cassette or LoxP as they have been removed.

    Phenotype
    The Chd5 gene is knocked out resulting Chd5 KO males become infertile. The Chd5 heterozygous males and Chd5 KO females are unaffected. Some mice may develop spontaneous tumors later in life (unconfirmed observation).

    Strain Development
    To generate the Chd5 KO strain; 12 kb of the Chd5 locus spanning from intron 6 to intron 18 was cloned and manipulated to construct a targeting vector. LoxP sites were inserted at loxP1-GTCTCTAGATGAGGCTG and CCTGCCCAGCATCTAC- loxP2, and a PGK-neo cassette, with FRT sites on both ends, was inserted into the immediate 5’-region upstream of the 2nd LoxP site. Female mice with the targeted allele were mated with129S/Sv-Tg(Prm-cre)58Og/J male mice (JAX Strain #003328) to delete exons 12 and 13 in the male offspring. They were then mated with 129S2/SvPasCrl wild-type mice (129-E Charles River) to generate constitutive Chd5+/− mice. Chd5 heterozygous male and female mice were mated to generate Chd5 +/+, +/−, −/− mice. The resulting strain was then backcrossed to 129X1/SvJ for 10+ generations and colony maintained with sib-mating.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
