Strain Development

Tg(Fev-tTs)1Leod was produced by cloning the coding sequence of tTS protein followed by an SV40 polyadenylation signal (PMID:7891684) into the T3 polylinker region of the NarI/BgzA modification 5 plasmid, placing the coding sequence downstream of a b-globin promoter (PMID:16251278). The b-globin promoter and tTS coding sequence were then released with an RsrII digest and the resulting fragment was cloned into the EPet-1Mini-BACMod#1 containing a 40-kb genomic DNA fragment (540z) that lies upstream of the Fev coding sequence (PMID:16251278). The resulting BAC EPet-1-tTs was linearized and subjected to pronuclear injection into B6CBAF2 hybrid zygotes. Founders were identified by PCR. Animals harboring the Tg(Fev-tTs)1Leod allele as well as the Htr1atm3.1Rhn allele (Cre deleter background unknown) were generated by breeding Tg(Fev-tTs)1Leod mice onto the Htr1atm3.1Rhn background. Tg(Fev-tTs)1Leod/Htr1atm3.1Rhn males were then bred to Htr1atm3.1Rhn females. As a result, the Tg(Fev-tTs)1Leod transgene was transmitted through the male germline, ensuring that all pups were raised by mothers of the same genotype, regardless of doxycycline status. The line was backcrossed to the 129S6/SvEvTac more than four times. Upon arrival at the MMRRC, 129S1 oocytes were used, moving the genetic background designation to stock.
