The targeting construct "pDNDF-7-DTA-5’ arm-P2A-CreERT2-STOP-Frt-Neo-Frt-3’ arm-DTA" was linearized by BsiWI enzyme and then was electroporated into male C57BL/6N-derived JM8A3 ES cells. The JM8A3 ES cells were cultured as described PMID:19525957. ES cell colonies were selected by G418 (Sigma-Aldrich) for 7 days. Successful recombination was validated by Southern blot. Two positive ES cell clones were microinjected into blastocysts in the NCI (National Cancer Institute) Transgenic Mouse Facility. Male chimeric mice expressing more than 50% of agouti coat color were then bred with B6N.Cg-Tg(ACTFLPe)9205Dym/CjDswJ females (RRID:IMSR_JAX:019100).

The deletion of the Neo-Frt cassette and the residual Frt was confirmed by genotyping with primers Ald1-delNeo-F and Ald1-delNeo-R. The Aldh1a1-P2A-CreERT2/ACTBFLPe mice were backcrossed with wild-type C57BL/6J mice (RRID:IMSR_JAX:000664) to breed out the ACTBFLPe transgene. Aldh1a1-P2A-CerERT2 mice were further backcrossed with C57BL6/J mice for 5+ generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
